Increasing neuronal glucose uptake attenuates brain aging and promotes life span under dietary restriction in Drosophila

چکیده
Summary Brain neurons play a central role in organismal aging, but there is conflicting evidence about the of neuronal glucose availability because uptake and metabolism are associated with both aging extended life span. Here, we analyzed metabolic changes brain Drosophila during aging. Using genetically encoded fluorescent adenosine triphosphate (ATP) biosensor, found decreased ATP concentration somata aged flies, correlated content, expression transporter glycolytic enzymes mitochondrial quality. The age-associated reduction did not occur suppressed glycolysis or enhanced uptake, suggesting these pathways contribute to reductions. Despite damage, increasing maintained levels, locomotor deficits, Increasing dietary restriction resulted longest spans, an additive effect enhancing bioenergetic challenge on

Calories Do Not Explain Extension of Life Span by Dietary Restriction in Drosophila
Dietary restriction (DR) extends life span in diverse organisms, including mammals, and common mechanisms may be at work. DR is often known as calorie restriction, because it has been suggested that reduction of calories, rather than of particular nutrients in the diet, mediates extension of life span in rodents. We here demonstrate that extension of life span by DR in Drosophila is not attribu...
متن کاملDietary restriction delays aging, but not neuronal dysfunction, in Drosophila models of Alzheimer's disease
Dietary restriction (DR) extends lifespan in diverse organisms and, in animal and cellular models, can delay a range of aging-related diseases including Alzheimer's disease (AD). A better understanding of the mechanisms mediating these interactions, however, may reveal novel pathways involved in AD pathogenesis, and potential targets for disease-modifying treatments and biomarkers for disease p...
متن کاملDietary Restriction in Drosophila: Delayed Aging or Experimental Artefact?
Lifespan can be extended by reduction of dietary intake. This practice is referred to as dietary restriction (DR), and extension of lifespan by DR is evolutionarily conserved in taxonomically diverse organisms including yeast, invertebrates, and mammals. Although these two often-stated facts carry the implication that the mechanisms of DR are also evolutionarily conserved, extension of lifespan...
متن کاملAging, life span, and senescence.
Research in the field of aging recently has entered a new era. Model systems have begun to provide insight into cellular, molecular, and organismal changes that are related intimately to aging. One important approach has been the identification of genes that determine the life span of an organism. The very existence of genes that when mutated can extend life span suggests that one or a few proc...
متن کاملDietary Restriction and Nutrient Balance in Aging
Dietary regimens that favour reduced calorie intake delay aging and age-associated diseases. New evidences revealed that nutritional balance of dietary components without food restriction increases lifespan. Particular nutrients as several nitrogen sources, proteins, amino acid, and ammonium are implicated in life and healthspan regulation in different model organisms from yeast to mammals. Agi...
